Fundamentals of Oligonucleotide Separation by HPLC
July 15th 2020From siRNA to mRNA to DNA, separating full length product from impurities can be challenging. Taking into consideration the nature of the oligonucleotide you are interested in, you will be able to make informed choices about what approach to use to characterize or purify your molecule.
High-resolution Separation of Oligonucleotide using Anion Exchange HPLC columns
July 13th 2020Polymeric strong anion exchange methods such as PL SAX are suitable for anion exchange and purification of oligonucleotides. At elevated temperatures, enhanced mass transfer increases the resolution for larger oligonucleotides. Working at higher PH values enables resolution of phosphodiester and phosphothioate forms of the oligonucleotide.
