Accurate and sensitive all-ions quantification using ultra-high resolution LC-MS
Endogenous metabolite profiling is an extreme example of multi-component quantitative analysis. Triple quadrupole mass spectrometers are considered the gold standard for quantification using selected reaction monitoring (SRM). However, as the number of analytes increases, SRM method development becomes more labour intensive. Using shorter SRM dwell times to accommodate more components compromises sensitivity and precision. In contrast, ultra-high resolution LC–MS provides a highly sensitive and relatively unbiased quantitative assessment of metabolomics samples, without the need for MS–MS; it also obviates the need for multiple platforms for profiling and validation.
