The comprehensive profiling of oligo products is critical and several attributes including intact mass, purity, and sequence are routinely determined by high-performance liquid chromatography–mass spectrometry (LC–MS). A variety of instrument and software options for these examinations exist and it can be challenging to navigate between them. This presentation will focus on a range of LC–MS options for the characterization of oligo samples and the data analysis strategies behind those efforts. Specifically, oligo data from single quadrupole and multiple high-resolution accurate-mass instruments will be explored. In addition, software workflows for intact mass determination, purity assessment, and sequence confirmation will be demonstrated.
