LC-MS/MS Analysis of PFAS in Serum

Event Overview:
Per- and polyfluoroalkyl substances (PFAS) are exceptionally versatile chemicals and have seemingly endless applications. However, their persistent nature and prevalence in blood, food, and the environment, along with emerging concerns about their long-term toxicity, has led to efforts to detect and quantitate PFAS in serum and other biological samples. Analyzing PFAS is particularly challenging due to background contamination in laboratory supplies and systems. This webinar will cover sample preparation and LC-MS/MS analysis of an expanded panel of PFAS compounds in serum. We will present a microelution SPE method for 7 PFAS identified as significant by the National Academies of Sciences, Engineering, and Medicine for clinical research and biomonitoring in public health labs. Additionally, we will discuss strategies to reduce background contamination to improve limits of detection (LOD) and limits of quantification (LOQ), as well as LC separation techniques.

Shahana W. Huq
, MS, is a senior application scientist focusing on emerging technologies and solutions within clinical research and sample preparation at the Phenomenex applications laboratory since 2000. She holds a master of science degree in biochemistry from California State University, Los Angeles. Her work involves experimental design, analytical method development, optimization, and troubleshooting. Furthermore, she has extensive expertise in R&D product innovation and application development, as well as hands-on experience in sample preparation and LC-MS/MS. Huq’s drive and vision of the clinical field have motivated her to participate in various scientific publications and to coauthor a Phenomenex patent on sample preparation products. Currently, Huq works closely with global customers in the applied market, providing technical support and consultation services, as well as onsite seminars and webinars to promote peer development.

Richard Jack
, PhD, is the global market development manager for the environmental and food markets at Phenomenex. He has over 18 years of experience with chromatography and mass spectrometry for the environmental, food, cannabis, semiconductor, chemical, and pharmaceutical industries. Jack has collaborated with global regulatory agencies to develop validated methods through new applications, instrumentation, column chemistries, and software. He is a former EPA scientific advisor for the EPA’s panel on hydraulic fracturing and a coauthor for EPA 557 and 557.1, ASTM D8001, and updates to D4327 and D6919 methods. He is currently the second vice chairman for the ASTM D19 subcommittee on water analysis. Previously, Jack was a vertical marketing manager with Thermo Fisher Scientific, Dionex, and Hitachi High Technologies, where he designed analytical instrumentation, including IC and HPLC systems, pumps, autosamplers, and a variety of detectors. He received his PhD in biochemistry and anaerobic microbiology from Virginia Tech in Blacksburg, Virginia. He received his master’s degree in ecology from the University of Tennessee in Knoxville.
